Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3

Early human

Lipidated pentapeptide analogue(s)

Research material. Not for human or animal consumption.

Research overview

Palmitoyl Pentapeptide-4, sold as Matrixyl and formerly labelled Palmitoyl Pentapeptide-3, is the lipopeptide Pal-Lys-Thr-Thr-Lys-Ser (Pal-KTTKS). Its peptide portion is a fragment of the type I procollagen propeptide, and the palmitic acid tail was added to improve skin penetration. Cell work reported increased collagen and fibronectin synthesis in fibroblasts. Human evidence includes a 2005 double-blind split-face moisturiser study and a small 2023 randomised topical study, both reporting exploratory cosmetic signals.

Questions about Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3

PRODUCT QUESTIONS
What is Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3?

This listing covers palmitoylated KTTKS, a five residue peptide derived from type I procollagen and joined to palmitic acid. Palmitoyl pentapeptide 3 is an older INCI name for the same lipopeptide, which is now listed as palmitoyl pentapeptide 4.

What research has been published on Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3?

Fibroblast cultures exposed to KTTKS showed increased synthesis of collagen types I and III and fibronectin, which led to its use as a matrikine in skin care. Human data are small: a 2005 split face moisturiser trial and a 21 participant randomised study in 2023 reported changes in wrinkle and texture measures rather than therapeutic outcomes.

What batch documentation should accompany Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3?

Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3: Ask for the COA and analytical reports for the batch being quoted. The palmitoyl group makes the peptide poorly water soluble, so check that the HPLC method uses a suitable organic gradient, that mass spectrometry confirms the acylated mass, and that free KTTKS is reported as an impurity.

How should Palmitoyl Pentapeptide-4 / Palmitoyl Pentapeptide-3 storage conditions be assessed?

Keep this lipopeptide dry and sealed at 2°C to 8°C, or frozen for long storage, away from light. Its fatty acid chain encourages it to stick to plastics and form micelles, so solutions are usually prepared in glass with a cosolvent.
