Synthetic guanylate cyclase C agonist peptide

Research material. Not for human or animal consumption.

Research overview

Linaclotide is a 14-amino-acid peptide with three disulfide bonds, modelled on the heat-stable enterotoxins of E. coli and on the gut hormone guanylin. It activates guanylate cyclase-C on the intestinal lining, raising cyclic GMP. That drives chloride and bicarbonate secretion into the gut, softening stool, and reduces signalling in pain nerves. Taken by mouth, it acts within the gut with negligible absorption, and defined products (Linzess, Constella) are authorised for IBS with constipation and chronic idiopathic constipation.

Questions about Linaclotide

PRODUCT QUESTIONS
What is Linaclotide?

Linaclotide is a synthetic peptide agonist of guanylate cyclase C used in gastrointestinal research, notably in models of IBS C and chronic constipation.

What research has been published on Linaclotide?

Linaclotide activates guanylate cyclase C on the intestinal lining and increases epithelial cGMP signalling, which drives fluid secretion into the gut. Randomised trials of defined finished products reported improved bowel and abdominal symptom outcomes versus placebo in specified constipation related disorders. Authorisation, formulation, age limits and indications depend on the product and jurisdiction; this catalogue listing does not itself establish a clinically equivalent or authorised product.

What batch documentation should accompany Linaclotide?

Linaclotide: Ask for the COA and analytical reports for the batch being quoted. Correct pairing of its three disulfide bonds is essential, so ask for mass spectrometry, HPLC purity that resolves misfolded isomers and a confirmed disulfide pattern.

How should Linaclotide storage conditions be assessed?

Store Linaclotide dry and sealed, protected from moisture, as the finished capsules are. It is sensitive to humidity, so keep desiccant in storage containers and minimise opening time.
