Leuprorelin Acetate / Leuprolide Acetate

Authorised product

Synthetic GnRH agonist peptide

Research material. Not for human or animal consumption.

Research overview

Leuprorelin, known as leuprolide in the US, is a GnRH agonist with D-leucine at position 6 and an ethylamide C-terminus. It is one of the most widely used peptide medicines, mainly as monthly to six-monthly depot injections such as Lupron and Prostap. Continuous exposure first triggers a hormone flare, then shuts down pituitary LH and FSH release, lowering testosterone and oestrogen. Uses include advanced prostate cancer, endometriosis, uterine fibroids, central precocious puberty and some breast cancers.

Questions about Leuprorelin Acetate / Leuprolide Acetate

PRODUCT QUESTIONS
What is Leuprorelin Acetate / Leuprolide Acetate?

Leuprorelin Acetate / Leuprolide Acetate is the acetate salt of a nine residue GnRH analogue with D leucine at position 6 and an ethylamide at the C terminus. The two names describe the same substance.

What research has been published on Leuprorelin Acetate / Leuprolide Acetate?

Its clinical and regulatory history is extensive: testosterone suppression in prostate cancer, depot formulation studies using polymer microspheres and gels, endometriosis and fibroid trials, and puberty suppression in children. The authorised indications depend on formulation and jurisdiction.

What batch documentation should accompany Leuprorelin Acetate / Leuprolide Acetate?

Leuprorelin Acetate / Leuprolide Acetate: Ask for the COA and analytical reports for the batch being quoted. Ask for mass spectrometry, HPLC purity with the D serine and other racemisation products reported, chiral purity data and acetate content.

How should Leuprorelin Acetate / Leuprolide Acetate storage conditions be assessed?

Store the acetate salt dry at 2°C to 8°C or frozen, protected from light. Solutions are stable for some time when refrigerated, but avoid alkaline pH, which promotes racemisation.
