Larazotide Acetate
Late stageSynthetic peptide modulator of tight junctions
Research material. Not for human or animal consumption.
Research overview
Larazotide (AT-1001) is an eight-amino-acid peptide, Gly-Gly-Val-Leu-Val-Gln-Pro-Gly, derived from zonula occludens toxin made by Vibrio cholerae. It was designed to tighten the junctions between intestinal cells, which become leaky in coeliac disease when gluten triggers zonulin release. Taken by mouth before meals, it reached Phase 3 as an add-on to a gluten-free diet, but the trial was stopped in 2022 after an interim analysis suggested it was unlikely to meet its goal.
Substantial controlled clinical research or late stage development has been identified; this is not a product authorisation claim.
Questions about Larazotide Acetate
PRODUCT QUESTIONSWhat is Larazotide Acetate?
Larazotide Acetate is the acetate salt of an eight residue peptide derived from Vibrio cholerae zonula occludens toxin. It acts locally in the gut and is minimally absorbed.
What research has been published on Larazotide Acetate?
Phase 2 trials in coeliac disease measured symptoms during gluten challenge and on a gluten free diet, with some symptom improvements at lower doses. The Phase 3 CedLara trial was discontinued after interim results. Laboratory studies examine tight junction assembly in intestinal cell models.
What batch documentation should accompany Larazotide Acetate?
Larazotide Acetate: Ask for the COA and analytical reports for the batch being quoted. Ask for mass spectrometry, HPLC purity, peptide content and acetate content, with deamidated glutamine forms reported.
How should Larazotide Acetate storage conditions be assessed?
Store Larazotide Acetate dry at 2°C to 8°C or −20°C, protected from moisture. Its glutamine can deamidate slowly in solution, so prepare solutions fresh.
