Pasireotide
Synthetic somatostatin analogue
Research material. Not for human or animal consumption.
Research overview
Pasireotide (SOM230) is a cyclic hexapeptide somatostatin analogue. Earlier analogues such as octreotide bind mainly the SSTR2 receptor; pasireotide binds SSTR1, 2, 3 and especially SSTR5, which is abundant on corticotroph tumours. This broader profile is why it was developed for Cushing's disease, where it is authorised as Signifor, and for acromegaly as a long-acting formulation. Raised blood glucose is a characteristic effect, because SSTR5 activity reduces insulin and incretin secretion.
An authorisation applies to at least one defined finished product in a stated jurisdiction; it does not apply automatically to this catalogue material.
Questions about Pasireotide
PRODUCT QUESTIONSWhat is Pasireotide?
Pasireotide is a synthetic cyclohexapeptide containing unnatural amino acids that mimic the receptor binding loop of somatostatin. It is engineered to bind four of the five somatostatin receptor subtypes.
What research has been published on Pasireotide?
Phase III trials in Cushing's disease measured normalisation of urinary free cortisol, while acromegaly trials compared long acting pasireotide with octreotide and lanreotide on growth hormone and IGF 1 control. Other studies looked at neuroendocrine tumours and at reducing pancreatic fistula after surgery. Hyperglycaemia was the most consistent adverse finding.
What batch documentation should accompany Pasireotide?
Pasireotide: Ask for the COA and analytical reports for the batch being quoted. Expect mass spectrometry confirming the cyclic structure, HPLC purity with impurity limits, and a statement of which salt form, aspartate or pamoate, the batch represents.
How should Pasireotide storage conditions be assessed?
Keep lyophilised Pasireotide at 2°C to 8°C or frozen, protected from light and moisture. The salt form affects solubility, so check the supplier's data before preparing solutions.
