Synthetic peptide calcimimetic

Research material. Not for human or animal consumption.

Research overview

Etelcalcetide (Parsabiv) is a calcimimetic: it activates the calcium-sensing receptor on parathyroid cells, signalling them to release less parathyroid hormone. Unlike oral cinacalcet, it is a peptide given intravenously at the end of haemodialysis sessions. Structurally it is a chain of seven D-amino acids linked by a disulfide bond to an L-cysteine, and it binds the receptor through a covalent disulfide exchange. It is authorised for secondary hyperparathyroidism in adults on haemodialysis.

Questions about Etelcalcetide

PRODUCT QUESTIONS
What is Etelcalcetide?

Etelcalcetide is a synthetic peptide made of seven D amino acids, attached through a disulfide bond to a separate L cysteine. It is classed as a calcimimetic because it acts on the calcium sensing receptor.

What research has been published on Etelcalcetide?

Phase 3 placebo controlled trials in adults on haemodialysis measured the proportion achieving more than a 30 percent fall in parathyroid hormone, and a head to head trial compared it with cinacalcet. Studies also tracked serum calcium and phosphate and the risk of low calcium.

What batch documentation should accompany Etelcalcetide?

Etelcalcetide: Ask for the COA and analytical reports for the batch being quoted. Because the peptide is built from D amino acids and a disulfide link, the certificate should include chiral purity, mass spectrometry of the disulfide conjugate, HPLC purity and the hydrochloride salt content.

How should Etelcalcetide storage conditions be assessed?

Store Etelcalcetide at 2°C to 8°C, protected from light. Its disulfide link to cysteine can exchange with other thiols, so avoid reducing agents and thiol containing buffers.
