Somatostatin analogue peptide (radiolabelling precursor)

Research material. Not for human or animal consumption.

Research overview

Edotreotide, also called DOTATOC, is octreotide with tyrosine at position 3 and a DOTA chelator attached to its N-terminus. DOTA holds radioactive metals tightly, turning the somatostatin analogue into a targeting vehicle. Labelled with gallium-68, it is used in PET scans to locate somatostatin receptor-positive neuroendocrine tumours, an approach authorised in the US and Europe. Labelled with lutetium-177 or yttrium-90, it has been studied for peptide receptor radionuclide therapy.

Questions about Edotreotide

PRODUCT QUESTIONS
What is Edotreotide?

Edotreotide is a conjugate of the chelator DOTA and Tyr3 octreotide, a cyclic octapeptide with a disulfide bond. It is a precursor that must be labelled with a radiometal before use in imaging or therapy research.

What research has been published on Edotreotide?

Imaging studies compared gallium 68 DOTATOC PET with older octreotide scintigraphy and found better detection of neuroendocrine tumours. Therapy research with lutetium 177 or yttrium 90 labelled edotreotide measured tumour response and kidney exposure. Radiochemistry work optimised labelling yields and purity.

What batch documentation should accompany Edotreotide?

Edotreotide: Ask for the COA and analytical reports for the batch being quoted. For a radiolabelling precursor, ask for mass spectrometry, HPLC purity, free DOTA and unconjugated peptide levels, and trace metal content, because contaminating metals compete for the chelator.

How should Edotreotide storage conditions be assessed?

Store Edotreotide at −20°C, dry and sealed. Use metal free water and plasticware when preparing solutions, since trace metals bind the DOTA chelator and reduce later labelling yields.
