Cyclic RGD pentapeptide integrin inhibitor

Research material. Not for human or animal consumption.

Research overview

Cilengitide is the cyclic pentapeptide cyclo(Arg-Gly-Asp-D-Phe-N-Me-Val), designed by Horst Kessler's group as a selective inhibitor of the αvβ3 and αvβ5 integrins. These integrins help tumour and blood vessel cells attach to the surrounding matrix and are involved in tumour angiogenesis. Cilengitide reached Phase 3 in glioblastoma, but the CENTRIC trial, which added it to standard chemoradiotherapy in patients with methylated MGMT promoters, did not improve survival, and development stopped.

Questions about Cilengitide

PRODUCT QUESTIONS
What is Cilengitide?

Cilengitide is a cyclic five residue peptide built around the arginine, glycine, aspartate integrin recognition motif. A D phenylalanine and an N methylated valine lock it in a shape that favours alpha V integrins.

What research has been published on Cilengitide?

Preclinical studies showed reduced angiogenesis and tumour growth in glioma and other models. Clinical trials in glioblastoma, including CENTRIC and CORE, measured survival and found no benefit. It remains a reference compound in integrin and tumour vascular biology research.

What batch documentation should accompany Cilengitide?

Cilengitide: Ask for the COA and analytical reports for the batch being quoted. Ask for mass spectrometry confirming the cyclic structure, HPLC purity with the linear precursor reported, and chiral purity for the D phenylalanine.

How should Cilengitide storage conditions be assessed?

Cilengitide is a compact cyclic peptide and is relatively stable. Store it dry at 2°C to 8°C or −20°C, protected from moisture, and check solution stability for long experiments.
